hmmm is pas op 02-03 in de data base opgenomen en gala heeft het in de us in de avond goed gedaan, misschien was de opname in de database daar dus in de avond bekend en hier nog niet ( tijdens de handelsuren )

als dat zo is dan kan verklaart het waarom gala in de us toch mooi up ondanks dat de indexen flink rood gingen.

kunnen we morgen dik in het groen gaan zullen we zeggen een +3% tot 5% moet dan toch wel kunnen.

Hopelijk kan Galapagos snel vorderingen maken. Onno vertelde na de borrel dat ze platgebeld werden en patiënten zelfs naar Leiden komen om te vragen of ze aub aan een test mee mogen doen (zoiets hadden ze nog nooit meegemaakt).

Op GLPG site staat onder sec filings SC 13G/A An amendment to the SC 13G filing met Gilead van 8-3-2018. Ik vind dit altijd lastig te lezen, maar dit is in feite wat we in december toch al wisten (verschuiving naar Gilead Therapeutics A1 Unlimited Company, of lees ik dit verkeerd? Iemand?

Ik lees er hetzelfde in als jij; dit ziet toe op de verhanging van de aandelen GLPG in december. Het is wel vreemd dat dit nu pas wordt gedocumenteerd. Maar verder zie ik er geen additionele nieuwswaarde in.

updates Finch op clinical trails. Had gehoopt op volledige recruitment van Finch1 gezien de vorige update waarin heel veel centra op non-recruiting ging. Jammer, alleen 7 centra oid erbij. Maandje wachten maar weer.

Hier is alvast iemand die positief is. Een analyst van Leerink Partners. Jawel, de fameuze Geoffrey Porges.

Wat zijn mening al dan niet waard is, laat ik in het midden.

“The proof of this process is only the final regulatory decision, and historically the FDA’s caution in this indication may still limit the label, or dosing, granted to Xeljanz,” Porges cautioned clients.

Het unanieme positieve oordeel door het Gastrointestinal Drugs Advisory Committee van de FDA over Xeljanz kan ook gunstig zijn voor de andere JAK inhibitors Upadacitinib en Filgotinib:

Pfizer just got some backup on its quest to snag a nod for ulcerative colitis med Xeljanz.
On Thursday, the FDA’s GI drugs advisory committee voted unanimously to approve the drug, even recommending a green light for a higher 10-mg dose.
Regulators had expressed concerns about the larger dose’s safety, supporting only a 5-mg dose, Leerink Partners analyst Geoffrey Porges wrote in a Friday note to clients. But now, the vote “may force their hand in approving a broader label than we originally anticipated,” he said.
A 10-mg dose had been previously discontinued in both rheumatoid arthritis and psoriatic arthritis, Xeljanz’s other indications, because of serious side effects. But ulcerative colitis is notoriously tough to tackle, Porges pointed out, and Pfizer’s seeking the 10-mg approval only for the most difficult-to-treat patients.

De schuingedrukte zin is onjuist.
Deze studie (net als de meeste phase II en III studies) vind plaats op meerdere sites in verschillende landen. De proeven starten en eindigen op uiteenliggende tijden. Op sommige plekken zijn ze vaak al klaar als ze op andere nog moeten beginnen.

Zolang er op tenminste 1 locatie nog patienten worden gezocht (terwijl andere locaties al klaar zijn of vol zitten), blijft de status nog staan op "recruiting".

Als alle locaties vol zitten (of al klaar zijn), en dus de laatste patienten aan hun trial beginnen, verandert de status naar "active, not recruiting" . Vanaf dat punt kun je gaan schatten met de behandelduur tijdens de trial en de verzamel/verwerkingstijd daarna, wanneer de resultaten beschikbaar komen.

Als alle patienten het test traject doorlopen hebben, en daarna de uitslagen worden verzameld, veranderd de trial status tenslotte in "completed".

En aanvullend op Maxen kan je in de link van Zuiderbuur lezen dat

Estimated Enrollment : 1300 participants

Op basis van bovenstaande info zou je kunnen aannemen dat de laatste patiënt 10 weken voor November 2019 start. De 10 weekse data zou dan in November 2019 beschikbaar zijn. Echter, is het natuurlijk onzinnig om de beslissing om door te gaan in fase 3 pas bij de laatste patiënt te nemen. Deze beslissing wordt genomen over een subgroep van de patiënten die eerder is gestart.
Volgens de Gilead 4q17 rapportage, sheet 41, komt de interim futility analyse in 2q18. Als ik Galapagos , op de borrel, goed heb begrepen komt er alleen een doorgaan in fase 3 of stop beslissing. Onderliggende data zoals safety en efficiëntie worden niet gepubliceerd en blijven zelfs blinded voor Galapagos en Gilead. (klopt dit laatste ?)

November 2019 is ver weg. Zoek op dezelfde site eens de studie Finch 1 op (https://clinicaltrials.gov/ct2/show/NCT02889796). Deze study heeft een primary datum van April 2019 en dat is een 12 weeks score. Gilead heeft gecommuniceerd dat deze trail in 2q18 volledig gerecruiteerd is. Primary completion zal dus voor mid September 2018 zijn (ipv April 2019). Als je een minuut over hebt kan je bijna helemaal onderaan klikken op "history of changes". In het nieuwe window klik je dan op "Beta archive site". Als je dan in kolom A op bv nr 19 january 10 klikt en in B op nr 21 maart 9 klikt en dan compare kiest zal je zien dat er na 19 january heel veel sites op "active, not recruiting" zijn over gegaan. Er zijn nog maar een paar sites open (voor nieuwe patiënten). Gilead komt elke maand met een update van deze info, leuk om in de gaten te houden.

UC is een fase 2-3 omdat filgotinib voordien nog niet eerder in patienten getest is. Dus is er een interim analye van een aantal patienten, na 6 maaanden. Met een positief resultaat wordt de trial voortgezet en wordt het een fase 3. Galapagos, en wij dus ook, krijgen die data idd niet te zien, blijft dubble blind. Een aantal mensen van het monitoring committee zien de data. Dus als de trial doorgaat naar fase 3, wordt dit door Galapagos aangekondigd en kunnen we dit als heel goed nieuws beschouwen.

Vraag: ik lees dat Incyte een JAK1 selective inhibitor ontwikkelt (INCB0527903) gericht op Acute Myeloid Leukemia (AML). Iemand een idee of Filgotinib als JAK1 ook in dit veld een rol zou kunnen spelen, of is het slechts selectief op ontstekingsziekten?
