Goed dat BlueBird het gen-sentiment een impuls geeft.
41 USD genoteerd meer dan een miljoen aandelen handel: daar moet Uniqure ook naar toe.
Dankzij positieve ontwikkelingen in de sector hebben we toch alweer de 13 USD gezien afgelopen week ondanks einde Locked up begin augustus.
Hopelijk blijft dit sentiment in H2, draait deze positieve wind dan zal de 8 USD wel weer in zicht komen begin augustus.

Zoals u weet zijn de Amerikaanse indices nog altijd leidend in de wereld en kunnen de futs met minimale middelen worden beinvloed en daarmee de beurzen all over the world. Maar na 5 jaren van onafgebroken stijging, en zonder een echte correctie van 10-15%, is de kans hierop steeds groter aan het worden. Daarnaast worden de macro factoren voor een stijging steeds minder gunstig. Ook vanuit technisch oogpunt bezien acht ik de kans zeer groot op een daling de komende weken met een climax waarbij mijn koersdoel 1750 is voor de S&P 500.

Negatieve divergentie tussen de indices en in de indices zelf
Draghi effect uitgewerkt
Toenemende onrust in het Midden-Oosten
Veel IPO’s

VIX historisch laag
Stijging olieprijs rem op groei
Goud weer begonnen aan uptrend?

Dimension leaps into gene therapy action with $252M Bayer deal, $30M round

Over the past 7 months, Dimension Therapeutics has been moving fast to take its place among a new wave of gene therapy developers looking to revolutionize the way some diseases are treated. Seeded by Fidelity Biosciences and working with a delivery vehicle developed by a noted researcher in the field, the Cambridge, MA-based biotech will later today unveil a $252 million licensing deal with Bayer--complete with a $20 million upfront--and a $30 million Series A round put up by Fidelity and new investor OrbiMed.

In the deal with Bayer, which is using its new pact to jump back into gene therapy, the pharma giant will fund the remaining preclinical work being done at Dimension on a treatment for hemophilia A as well as a followup Phase I/IIa study. Bayer can then take over for Phase III, with worldwide commercialization rights and a royalty stream reserved for Dimension, if all goes well.

Dimension made the leap into gene therapy as a new generation of biotechs led by the likes of bluebird bio has been gaining headlines with some remarkable results on just a handful of patients. Gene therapies that target small patient populations can deliver proof of concept data with just a handful of patients. And late-stage testing can come quickly and be completed with relatively tiny groups, when you compare them to big diseases like diabetes or heart failure.

Gene therapy involves inserting corrective genes into malfunctioning cells to get them to work again without further use of drugs. Dimension has been looking into using viruses to carry the genes to the affected cells.Gene therapy has seen more than 20 years of experiments and a series of disappointments. However, Amsterdam-based UniQure is about to start selling its gene therapy drug Glybera in Europe as a treatment for the ultra-rare disease lipoprotein lipase deficiency (LPLD) with partner Chiesi.

U.S. biotech firm Bluebird Bio last week reported positive results with an experimental gene therapy for another blood disorder, sending its shares soaring.

Dimension Therapeutics will receive an upfront payment of $20 million and may get development and commercialisation milestone payments of up to $232 million. Bayer will also make royalty payments based on future product sales.

Standard haemophilia A therapies are often administered intravenously multiple times a week and may be required for life.

The German drugmaker's established haemophilia A therapy Kogenate had 1.2 billion euros ($1.6 billion) in sales last year. It has two more drug candidates against the type A of the hereditary bleeding disorder in the third and last phase of testing on humans that is required for regulatory approval. ($1 = 0.7366 Euros)

UniQure in maand juni mooi herstel van 57%. Denk vooral door positieve berichten vanuit Bluebird die gehele sector hoger hebben gezet. Kwam dit overzichtje tegen op Twitter waar koersverloop van verschillende "gene therapy" bedrijven over afgelopen maand wordt weergegeven.

New here, yes. Big believer in uniQure and bought shares in the $8s. Found this forum and have been translating from Dutch to English (don't speak Dutch). Glad someone speaks English here. What do you think of the video?

An outstanding video. Last years, news around uniQure has been very reluctant. Most of the news is sent via 'very brief' press releases. This video shows more details and even emotion. It's also the reason why I visit shareholders meetings of uniQure; to see the faces behind the technology.

As you'll probably know, uniQure recently presented strong data concerning successful transfection of liver cells and concurrent safety data of its AIP trial, which is also displayed in the video. Although clinical data on the efficacy of the AIP gene therapy are expected at the end of 2014, the importance of this video is that it suggests a promising clinical response in at least one of the eight patients treated. For me that's new information. I hope/assume that the clinical results in the 7 remaining patients will demonstrate a similar effect.

By the way, if you read the written story under the video, look at what the third paragraph says:

"During the phase I of a clinical trial Angeles and seven other patients have received this treatment, and so far AIP attacks have been reduced significantly among this group. In addition, there are no side effects visible. These promising results indicate that within a few years, the new therapy could be available to all patients."

It sounds like ALL 8 patients are seeing positive clinical responses

What matters is the relationship between dose (gc/kg) and degree of (positive) expression (and to which degree the results are permanent).

The video gives a hopeful impression, but perhaps she (the woman in the video) received the highest dose. Others with a lower dose might benefit less (or not at all). We have to wait for the full dataset before drawing conclusions.

Vaak wordt getracht voor elke keer dat iets ingebracht moet worden, een vector te creëren waarop het immuunsysteem niet reageert. Een volgende poging wordt vaak door de inmiddels ontwikkelde afweer ongedaan gemaakt.

Misschien vindt men ooit een manier (geen idee of dat al getracht is; ben geen medicus/biotechnicus) om de immuunresponse/afweer tijdelijk/plaatselijk uit te schakelen?

De theorie is natuurlijk dat er slechts een éénmalige behandeling nodig is die hopelijk een leven lang mee gaat. Voorlopig moeten patienten zich troosten met het gegeven dat het leven na de behandeling kwalitatief een stuk is verbeterd.

Ik fantaseer verder: misschien wordt er ooit iets bedacht, waardoor het lichaam zelf in staat zal zijn om de ingebrachte goedwerkende genen, zelf te kopiëren/creëren...zodat herhaling van een behandeling niet nodig is. Ik ken dat wel van iPS cell technology (stamcellen), waar het bedrijf Cellectis onlangs de grootste deal in de franse biotech historie heeft gesloten met Pfizer.

Het is een speelbal, positieve ontwikkelingen in de pijplijn zijn broodnodig alhier !

Kijkend naar het koersverloop de afgelopen tijd is het blijkbaar heel eenvoudig om de koers te beinvloeden met relatief kleine volumes (vandaag weer 56K) Als middel tot grote partijen een koersverandering teweeg willen brengen is dat kinderspel.
